Antigen:

Calcitonin gene-related peptide is a 37 amino-acid vasoactive neuropeptide (mw 3789) widely expressed throughout the central and peripheral nervous system. CGRP has potent vasodilatory effects probably serving as a regulator of regional blood flow, in addition to a variety of other cellular effects.

Synthetic human alfa-CGRP coupled to carrier

Specificity:

ABS 026-05 binds human alfa-CGRP. Based on inhibition ELISA, ABS 026-05 shows full cross-reactivity with rat alfa-CGRP and about about 50% cross-reactivity with human beta-CGRP. Cross-reactivity with coated rat alfa-CGRP is low (8%) and there is no cross-reaction with human amylin.

Epitope specificity:

The epitope lies within residues 1-18 of human alfa-CGRP

Reactivity:

ABS 026-05 binds free alfa-CGRP in solution. Based on inhibition ELISA, the affinity of ABS 026-05 is at least 24 times higher than that of ABS 026-04.
The ELISA dilution guideline of applies to human alfa-CGRP coated directly at 1.0 g/mL. showing that the antibody binds poorly to the denatured peptide.

ABS 026-05 (as capture antibody) forms a sandwich ELISA pair with ABS 026-04 (as biotinylated detection antibody) for measuring human alfa-CGRP, giving a detection limit of 0.13 nmol/L in an unoptimized buffer assay.
